目的:研究丹参酮ⅡA磺酸钠(sodium tanshinoneⅡ-A sulfonate,DS-201)对大鼠骨髓间充质干细胞(Bone marrow stromal cells,BMSCs)体外增殖的影响。方法:对大鼠骨髓间充质干细胞进行体外培养和纯化扩增,取第3代细胞鉴定后进行实验。观察细胞在培养传代过程中的形态学变化;用MTT法筛选24h内对BMSCs生长最适的DS-201浓度,并用此浓度继续干预BMSCs生长;检测对照组和DS-201最适浓度组连续4天的吸光光度值,计算增殖率。结果:1.5×103μmol/LDS-201对BMSCs增殖有明显的抑制作用,在1.5×10-4~1.5×102μmol/L浓度范围内DS-201对BMSCs有促进增殖作用。其中1.5×10-2μmol/L组促增殖作用最大,并且在作用24h后的增殖率最高。但随着时间的延长此浓度的增殖率逐渐下降。结论:适宜浓度以及一定的作用时间内,DS-201对BMSC具有促增殖作用。
Objective: To study the effect of sodium tanshinoneⅡ-A sulfonate (DS-201) on the proliferation of Bone marrow stromal cells (BMSCs) in vitro. Methods: Rat bone marrow mesenchymal stem cells were cultured in vitro and purified. The cells were identified by passage 3 and then tested. The morphological changes of the cells during the passage of culture were observed. The optimal concentration of DS-201 for BMSCs was screened by MTT method and the growth of BMSCs was continued with this concentration. The optimal concentrations of DS-201 and control group Day absorbance value, calculate the proliferation rate. Results: The proliferation of BMSCs was significantly inhibited by 1.5 × 103μmol / L DS-201. The proliferation of BMSCs was promoted by DS-201 in the concentration range of 1.5 × 10-4 ~ 1.5 × 102μmol / L. Among them, 1.5 × 10-2μmol / L group had the highest proliferative effect and the highest proliferation rate after 24h. However, with the extension of time, the proliferation rate of this concentration gradually decreased. Conclusion: DS-201 can promote the proliferation of BMSC in proper concentration and for a certain period of action.
