为了提高羊肚菌胞外多糖得率,对羊肚菌液体培养进行了研究。通过单因素实验确定了最优碳源为玉米粉,最优氮源为蛋白胨,最佳生长因子为麸皮;采用正交实验,确定了羊肚菌发酵的最优培养基配方为:玉米粉3%,蛋白胨0.3%,麸皮2%,MgSO40.2%,KH2PO40.2%;并采用二次回归正交组合设计确定了最优培养条件为:温度25℃,摇床转速96r/min。在发酵罐扩大实验中,对胞外多糖含量进行了监测,其中发酵108h时胞外多糖含量达1.145g/L。
In order to improve the yield of Morchella extracellular polysaccharide, Morchella liquid culture was studied. The best carbon source was corn flour, the best nitrogen source was peptone and the best growth factor was bran by single factor experiment. The optimum medium for the fermentation of Morchella was determined by orthogonal experiment: corn flour 3%, peptone 0.3%, bran 2%, MgSO440.2% and KH2PO40.2%. The orthogonal design was used to determine the optimum culture conditions: temperature 25 ℃, shaking speed 96r / min. In the fermentation tank expansion experiment, the content of extracellular polysaccharide was monitored, in which the content of extracellular polysaccharide reached 1.145g / L at 108h.
