目的建立2型糖尿病性心肌病大鼠模型,探讨糖尿病引起心肌损伤的相关因素。方法 SD大鼠喂以高脂高糖饲料加小剂量链脲佐菌素,建立2型糖尿病胰岛素抵抗大鼠模型,随机分为正常组、糖尿病组。检测了两组大鼠血清中相关指标含量以及心肌组织中晚期糖基化终末产物(AGEs)、肿瘤坏死因子-α(TNF-α)、血管紧张素Ⅱ(AngⅡ)的含量,并观察心脏结构变化。结果糖尿病组相关指标均较正常组明显增高,差异有统计学意义(P<0.01),同时HE染色观察到糖尿病大鼠心肌细胞变性坏死,结构紊乱。结论糖尿病引起心肌病变的原因可能是细胞代谢紊乱引起心肌组织相关指标过度表达有关。
Objective To establish a rat model of type 2 diabetic cardiomyopathy and investigate the related factors of myocardial injury caused by diabetes mellitus. Methods SD rats were fed with high-fat and high-sugar diet plus a small dose of streptozotocin to establish a rat model of type 2 diabetic insulin resistance and were randomly divided into normal group and diabetic group. The serum levels of relevant indicators and the levels of advanced glycosylation end products (AGEs), tumor necrosis factor-α (TNF-α) and angiotensin Ⅱ (AngⅡ) Structural changes. Results Compared with the normal control group, the indexes in diabetic group were significantly higher than those in normal group (P <0.01). At the same time, the myocardial cells of diabetic rats were observed to be degenerated and necrotic, with structural disorder. Conclusions The possible cause of diabetic cardiomyopathy may be related to the over-expression of myocardial tissue-related index caused by disorder of cell metabolism.
