目的探讨大黄素对内毒素激活后巨噬细胞高迁移率族蛋白B1(HMGB1)基因表达和胞外释放的影响。方法内毒素脂多糖激活后小鼠腹腔巨噬细胞培养物经不同浓度(5、20、50μmol/L)大黄素作用一定时间,观察培养上清液HMGB1、TNF-α含量的变化及细胞HMGB1mRNA表达的改变。结果大黄素(50μmol/L)明显抑制脂多糖激活后小鼠腹腔巨噬细胞HMGB1mRNA的表达和HMGB1、TNF-α的释放(均P(0.01)。结论大黄素具有抑制巨噬细胞HMGB1mRNA表达和HMGB1胞外释放的作用。
Objective To investigate the effect of emodin on the gene expression and extracellular release of high mobility group box 1 (HMGB1) macrophage activated by endotoxin. Methods The peritoneal macrophages cultures of endotoxin induced by lipopolysaccharide (LPS) were treated with emodin at different concentrations (5, 20 and 50 μmol / L) for a period of time. The contents of HMGB1 and TNF-α and the expressions of HMGB1 mRNA Change. Results Emodin (50 μmol / L) significantly inhibited HMGB1 mRNA and HMGB1, TNF-α release in peritoneal macrophages (P <0.01) .Conclusion Emodin can inhibit the expression of HMGB1 mRNA and HMGB1 Extracellular release of the role.
