目的建立反相高效液相色谱法测定人血浆中奥卡西平浓度的方法,为临床合理用药提供依据。方法血浆蛋白经三氯乙酸处理后,采取峰面积外标法,用symmetryC18反相色谱柱,柱温25℃,以乙腈-水(30∶70)为流动相,流速1.0 ml/min,检测波长230 nm。结果血浆中奥卡西平在1.1μg/ml~275μg/ml的浓度范围内线性关系良好,回归方程:Y=56900C+76300.R=0.9998。结论本方法专属性强,灵敏度高,准确度高,线性范围宽,操作简便,分析快速,可用于奥卡西平血药浓度的检测。
Objective To establish a method for the determination of oxcarbazepine in human plasma by reversed-phase high performance liquid chromatography (RP-HPLC), and provide the basis for clinical rational drug use. Methods After the plasma protein was treated with trichloroacetic acid, the peak area external standard method was used. The column temperature was 25 ℃ and acetonitrile / water (30:70) was used as the mobile phase at a flow rate of 1.0 ml / min with a symmetry C18 reversed-phase column. The detection wavelength 230 nm. Results The concentration of oxcarbazepine in plasma ranged from 1.1μg / ml to 275μg / ml. The regression equation was Y = 56900C + 76300.R = 0.9998. Conclusion This method is highly specific, sensitive, accurate, linear range, easy to operate, rapid analysis, can be used for the determination of oxcarbazepine concentration.
