【目的】为了评估Vsp I,Sty I和Stu I为基础的线粒体细胞色素氧化酶I基因(mt COI)酶切扩增多态性序列(cleaved amplified polymorphic sequence,CAPS)标记方法鉴别烟粉虱Q与B隐种的有效性。【方法】本研究对国内外烟粉虱种群的mt COI基因进行了测序并鉴定了其隐种;在对464个Q隐种、98个B隐种mt COI序列分析的基础上,利用Vsp I,Sty I和Stu I对Q与B隐种分别进行了CAPS标记验证。检索并比对了Gen Bank中烟粉虱Q与B隐种mt COI序列中Vsp I,Sty I和Stu I酶切位点分布情况。【结果】对国内外烟粉虱种群研究发现,以Vsp I为基础的CAPS标记方法能够有效鉴别实验中的Q与B隐种;利用Stu I或Sty I的CAPS标记方法无法有效鉴别Q与B隐种。对Gen Bank中烟粉虱Q与B隐种mt COI序列比对发现,利用Vsp I,Sty I和Stu I为基础的CAPS标记方法不能有效鉴别Q与B隐种。【结论】以Vsp I,Sty I和Stu I为基础的CAPS标记方法鉴别Q与B隐种方面均有一定的局限性。
【Objective】 To identify the nucleotide sequence of mitochondrial cytochrome oxidase I (mt COI) cleavage amplified polymorphic sequence (CAPS) based on Vsp I, Sty I and Stu I Effectiveness with B occultation. 【Method】 The mt COI gene of Bemisia tabaci (B. tabaci) populations at home and abroad was sequenced and its hidden species were identified. Based on the mt COI sequence analysis of 464 Q hidden species and 98 B occult species, , Sty I and Stu I validated CAPS markers for Q and B occultations, respectively. The distribution of Vsp I, Sty I and Stu I in mt COI sequences of Q and B occidentalis of Bemisia tabaci GenBank was searched and compared. 【Result】 The study on Bemisia tabaci populations at home and abroad found that the CAPS labeling method based on Vsp I can effectively identify the Q and B occultations in the experiment. The CAPS labeling method using Stu I or Sty I can not effectively distinguish between Q and B Hidden species. The alignment of mt COI sequences between B. tabaci Q and B occultata in Gen Bank showed that the CAPS markers based on Vsp I, Sty I and Stu I could not discriminate between Q and B subtypes. 【Conclusion】 The CAPS labeling method based on Vsp I, Sty I and Stu I has some limitations in identifying Q and B occult.
