目的从转录水平探讨大黄灵仙胶囊防治胆结石的可能机制。方法将60只普通豚鼠随机分为3组,每组20只。模型组和大黄灵仙胶囊治疗组采用致石饲料诱发胆囊结石。治疗组造模开始即分别按剂量予大黄灵仙胶囊混悬液灌胃,模型组和正常对照组以等量生理盐水灌胃。8周后统计胆囊结石形成率;运用实时荧光定量PCR方法检测各组豚鼠肝组织胆固醇7α一羟化酶(CYP7A1)mRNA的表达情况。结果模型组、治疗组豚鼠的胆囊结石形成率分别为100%和50%,与模型组相比,治疗组结石形成率明显降低(P<0.01)。模型组豚鼠肝组织中的CYP7A1mRNA表达较正常对照组显著减少(P<0.01),而治疗组豚鼠肝组织中的CYP7A1mRNA表达是模型组的2.13倍,差异有统计学意义(P<0.01)。结论大黄灵仙胶囊具有较好的预防结石形成的作用,其机制可能与其具有减轻肝细胞损伤、间接稳定CYP7A1mRNA表达的作用有关。
Objective To explore the possible mechanism of rhubarb Lingxian capsule in preventing and treating gallstones from the transcriptional level. Methods Sixty normal guinea pigs were randomly divided into three groups of 20 rats. The model group and the rhubarb and Lingxian capsule treatment group induced the gallstones by inducing stone feeding. At the beginning of treatment, the rats in the treatment group were dosed with rhubarb Lingxian capsule suspension respectively, and the model group and the normal control group were given gavage with normal saline. The incidence of gallstone formation was measured after 8 weeks. The mRNA expression of cholesterol 7α-hydroxylase (CYP7A1) in liver of each group was detected by real-time fluorescence quantitative PCR. Results The formation rate of gallstone in model group and treatment group was 100% and 50%, respectively. Compared with model group, the formation rate of stone in treatment group was significantly lower (P <0.01). The expression of CYP7A1mRNA in liver tissue of model group was significantly lower than that of normal control group (P <0.01), while the expression of CYP7A1mRNA in liver tissue of model group was 2.13 times of the model group (P <0.01). Conclusion Rhubarb Lingxian Capsule has a good effect in preventing the formation of stones, and its mechanism may be related to its role in reducing hepatocellular injury and indirectly stabilizing CYP7A1 mRNA expression.
