目的:探讨硫酸右旋糖苷(DS)通过调节HIF-1α(hypoxia induced factor,HIF-1α)的蛋白表达而抑制人胃癌细胞腹膜转移的相关机制。方法:(1)体外实验:培养BGC-823细胞,分别在培养液中加入DS和PBS,低氧培养不同时间,用免疫细胞化学和WB检测HIF-1α表达。(2)动物实验:构建胃癌腹膜转移模型,实验组与对照组分别注射DS和PBS。剖腹观察癌细胞在腹膜上种植转移情况;应用免疫组化和WB方法检测HIF-1α表达。结果:(1)体外实验:免疫细胞化学和WB结果显示实验组HIF-1α表达明显低于对照组。(2)动物实验:实验组瘤结节数明显少于对照组,免疫组化和WB结果显示实验组HIF-1α的蛋白表达明显低于对照组。结论:硫酸右旋糖苷可以调节人胃癌细胞HIF-1α的蛋白表达,其可能会通过下调HIF-1α的表达从而抑制人胃癌细胞大网膜种植转移。
Objective: To investigate the mechanism of dextran sulfate (DS) inhibiting peritoneal metastasis of human gastric cancer cells by regulating the expression of HIF-1α protein. Methods: (1) In vitro experiments: BGC-823 cells were cultured, DS and PBS were respectively added to the culture medium and cultured in hypoxia for different time. The expression of HIF-1α was detected by immunocytochemistry and WB. (2) Animal experiment: The model of peritoneal metastasis of gastric cancer was constructed. The experimental group and the control group were injected with DS and PBS respectively. The laparotomy was used to observe the metastasis of cancer cells on the peritoneum. The expression of HIF-1α was detected by immunohistochemistry and WB methods. Results: (1) In vitro experiments: Immunocytochemistry and WB results showed that the expression of HIF-1α in experimental group was significantly lower than that in control group. (2) Animal experiments: The number of tumor nodules in the experimental group was significantly less than that in the control group, and the immunohistochemistry and WB results showed that the protein expression of HIF-1α in the experimental group was significantly lower than that in the control group. Conclusion: Dextran sulfate can regulate the protein expression of HIF-1α in human gastric cancer cells, which may inhibit the omentum metastasis of human gastric cancer cells by down-regulating the expression of HIF-1α.
