目的建立一种简易的疟色素提取方法,提高拉曼光谱技术检测疟疾的灵敏度。方法利用皂素的破膜特性提取疟色素,采用显微共聚焦拉曼光谱仪进行检测,通过软件处理拉曼信号,用化学计量法比对分析。结果疟色素提取物与阳性对照的拉曼光谱共有峰的相对位移频率、相对峰强度具有较好的一致性,与阴性对照没有相关性,检测的最小虫血率为0.1%。结论皂素可用于疟色素提取。疟色素提取物拉曼光谱检测能有效区分疟原虫感染红细胞与正常红细胞。
Objective To establish a simple method for extracting malaria pigment and improve the sensitivity of Raman spectroscopy in detecting malaria. Methods The malaria pigment was extracted from the rupture of saponin and detected by microscopy confocal Raman spectroscopy. The Raman signal was processed by software and compared with stoichiometry. Results The relative peak shift frequency and relative peak intensity of the common peaks of the malachite extract and the positive control were in good agreement. There was no correlation with the negative control, and the minimum detection rate was 0.1%. Conclusion Saponin can be used for malaria pigment extraction. Raman spectroscopy of malaria pigment extract can effectively distinguish between erythrocytes and normal erythrocytes infected by Plasmodium.
