目的探讨卡介苗多糖核酸(BCG-PSN)对口腔扁平苔藓(OLP)病损区培养角质细胞分泌细胞因子IL-12p40的影响和意义。方法取经病理诊断为口腔扁平苔藓病损组织做细胞培养:分为OLP组、OLP+BCG-PSN组,每组6例,在角质形成细胞长满时加入BCG-PSN,用同等体积生理盐水作为阴性对照,在培养6h、12h、24h、48h分别收集培养细胞上清液,ELISA法检测IL-12p40浓度变化情况。结果 1整个细胞生长期间为单一的角质形成细胞,细胞为椭圆形或多角形,呈现典型的铺路石状,符合上皮角质形成细胞生长形态特征。2在不同的培养时段内,BCG-PSN共同培养刺激角质形成细胞产生IL-12p40的减少量与对照组相比均有显著差异(6h F=7.87 P=0.0186,P<0.05;12h F=10.31 P=0.0093 P<0.05;24h F=16.57 P=0.0022 P<0.05;48h F=17.3 P=0.002 P<0.05)。结论 BCGPSN可能通过对OLP角质形成细胞分泌IL-12p40有直接的抑制效果起到减轻T细胞引起的直接损伤、降低局部自身免疫反应的作用。
Objective To investigate the effect of BCG-PSN on the secretion of cytokine IL-12p40 from keratinocytes cultured in the lesion of oral lichen planus (OLP). Methods Pathological diagnosis of oral lichen planus lesions tissue culture: divided into OLP group, OLP + BCG-PSN group, 6 cases in each group, added BCG-PSN when keratinocytes overgrowth, with the same volume of saline as Negative control, culture supernatants were collected at 6h, 12h, 24h and 48h respectively. The concentration of IL-12p40 was detected by ELISA. Results 1 The whole cell was single keratinocytes during the growth of the cells. The cells were oval or polygonal, showing a typical paving stone, which was consistent with the morphological features of epithelial keratinocytes. 2 In the different culture periods, the reduction of IL-12p40 induced by BCG-PSN co-cultured keratinocytes was significantly different from that of the control group (6h F = 7.87 P = 0.0186, P <0.05; 12h F = 10.31 P = 0.0093 P <0.05; 24h F = 16.57 P = 0.0022 P <0.05; 48h F = 17.3 P = 0.002 P <0.05). Conclusions BCGPSN may play a direct role in inhibiting the secretion of IL-12p40 by OLP keratinocytes and reduce the direct damage caused by T cells and decrease the local autoimmune response.
