应用离子交换层析法,从胎脑组织匀浆上清中提纯神经生长因子(nervesrowthfactor,NGF),其分子量约14.3KD,其3种组分的等电点分别为9.13、9.42和9.65。纯化后的NGF比活力显著提高,用鸡胚背根神经书培养法证明其具有刺激神经生长的作用。采取肌肉、球后和局部注射的方法,对45例病人进行了治疗观察。其中脑梗塞10例,脑外伤10例,外周神经损伤8例,青光眼14例,椎间盘脱出3例。总有效率为100%。NGF对大多数神经损伤表现均有作用,尤对弥漫性脑损伤、青光眼、外周神经损伤疗效最明显,未发现不良反应。
Nerve growth factor (NGF) was purified from fetal brain tissue homogenate by ion exchange chromatography and its molecular weight was about 14.3KD. The isoelectric points of the three components were 9.13,9. 42 and 9.65. The specific activity of purified NGF was significantly increased, which was proved by chick embryo nerve root book culture method to stimulate nerve growth. Forty five patients were treated with muscle, ball and local injection. Including cerebral infarction in 10 cases, 10 cases of traumatic brain injury, peripheral nerve injury in 8 cases, 14 cases of glaucoma, disc herniation in 3 cases. The total effective rate is 100%. NGF plays a role in most nerve injury, especially for diffuse brain injury, glaucoma and peripheral nerve injury. No adverse reaction was found.
