目的非甾体类抗感染药(nonsteroid anti-inflammatory drugs,NSAIDs)以其抗感染和抗风湿的作用广泛应用于临床,近年来,NSAIDs对多种肿瘤的防治作用被广泛关注。本研究探讨NSAIDs阿司匹林(aspirin,ASP)和吲哚美辛(indomethacin,Indo)抑制胃癌细胞增殖的作用和机制。方法 MTT法检测2、4、8、10mmol/L ASP以及200、400、800、1 000μmol/L Indo对MGC803细胞增殖活性的影响;蛋白质印迹法检测ASP和Indo对MGC803细胞中CRP和IL-6蛋白表达的影响。结果不同浓度的ASP和Indo对MGC803细胞具有增殖抑制作用,其中8 mmol/L ASP组和800μmol/L Indo组细胞的增殖抑制最明显,8mmol/L ASP抑制率为(70.5±1.3)%,与2、4mmol/L的(18.8±1.1)%和(59.5±1.7)%比较,差异有统计学意义,P<0.01;800μmol/L Indo抑制率为(79.9±1.9)%,与200、400、1 000μmol/L的(31.5±0.7)%、(49.9±1.6)%和(59.0±1.0)%比较,差异有统计学意义,P<0.01。并且8mmol/L ASP组和800μmol/L Indo组对MGC803细胞的增殖抑制还具有时间效应,作用48h的抑制效果明显,8mmol/L ASP48h抑制率为(83.9±1.6)%,与24h的(69.5±2.1)%比较,差异有统计学意义,P<0.05;800μmol/L Indo 48h抑制率为(82.4±1.4)%,与24、72、96h的(71.8±1.6)%、(72.8±1.6)%和(70.0±1.8)%比较,差异有统计学意义,P<0.05。MGC803细胞高表达CRP和IL-6,并且ASP和Indo能下调MGC803细胞CRP和IL-6蛋白的表达水平,8mmol/L ASP、800μmol/L Indo下调CRP的表达分别达79.0%和88.9%,下调IL-6的表达分别达49.8%和61.0%。结论 NSAIDs ASP和Indo对胃癌MGC803细胞具有增殖抑制作用,并能下调MGC803细胞中CRP和IL-6的表达。
The purpose of nonsteroidal anti-inflammatory drugs (NSAIDs) for its anti-infective and anti-rheumatic effects widely used in clinical practice, in recent years, NSAIDs on the prevention and treatment of many tumors has been widely concerned. This study was designed to investigate the effects and mechanisms of NSAIDs aspirin (ASP) and indomethacin (Indo) on gastric cancer cell proliferation. The down-regulation of CRP and down-regulation of CRP was found in 8mmol / L ASP and 800μmol / L Indo The expression of IL-6 reached 49.8% and 61.0% respectively. Conclusion NSAIDs ASP and Indo can inhibit the proliferation of gastric cancer MGC803 cells and down-regulate the expression of CRP and IL-6 in MGC803 cells.
