目的:探讨清肝活血方及其拆方对酒精性肝病(alcoholic liver d isease,ALD)大鼠血浆内毒素水平及KC活化状态的影响。方法:雄性W istar大鼠100只,SPF级,随机分成空白组、CC l4组各10只,余80只为造模组,采用复合因素复制ALD模型6周。空白组予等量生理盐水对照。CC l4组予微量CC l4腹腔注射2次/周。造模4周后将模型组随机分成4组,清肝活血方及其拆方组各15只,余为模型组。予等效剂量清肝活血方及拆方灌胃2周。检测血清ALT、AST水平;留取肝脏标本进行HE染色;采用偶氮显色法检测血浆内毒素水平;免疫组织化学染色检测肝组织CD68表达。结果采用SPSS12.0统计软件进行单因素方差分析及两两比较。结果:(1)清肝活血方及其拆方均可明显改善ALD大鼠肝脏脂肪变及肝脏炎症程度,清肝活血方>活血方>清肝方,清肝活血方优于清肝方。(2)清肝活血方能显著降低模型大鼠血清ALT水平,清肝方和活血方无明显作用;清肝方、活血方、清肝活血方均可明显降低ALD大鼠血清AST水平,组间比较无显著差异。(3)清肝活血方及其拆方均能降低模型大鼠血浆内毒素水平,组间比较无显著差异。(4)清肝活血方及其拆方均可抑制模型大鼠KC活化,活血方优于清肝活血方,清肝活血方优于清肝方。结论:清肝活血方及其拆方发挥对ALD的防治作用,其作用机理可能与降低血浆内毒素水平和抑制KC的活化有关。
Objective: To investigate the effects of Qinggan Huoxue Decoction and its decomposed prescriptions on plasma endotoxin levels and KC activation in alcoholic liver disease (ALD) rats. METHODS: A total of 100 male Wastar male rats were randomly divided into blank group and CC l4 group with 10 rats in each group. The remaining 80 rats were used as model group. The ALD model was duplicated for 6 weeks using compound factors. The blank group was given equal amount of saline control. The CC l4 group was given intraperitoneal injection of CC l4 twice a week. After 4 weeks of modeling, the model group was randomly divided into 4 groups, 15 each of Qinggan Huoxue Fang and its decombination group, and the remainder was the model group. The equivalent dose of Liver Activating Blood Circulation and disassembled prescriptions were given orally for 2 weeks. Serum ALT, AST levels were detected; liver specimens were obtained for HE staining; plasma azotoxin assay was used to detect plasma endotoxin levels; and immunohistochemical staining was used to detect CD68 expression in liver tissues. Results One-way ANOVA and pairwise comparison were performed using SPSS12.0 statistical software. Results: (1) Qinggan Huoxue Decoction and its decomposed prescriptions can obviously improve the hepatic steatosis and hepatic inflammation of ALD rats. Qinggan Huoxue Fang> Huoxue Fang> Qinggan Fang, Qinggan Huoxue Fang is better than Qinggan Fang. (2) Qinggan Huoxue can significantly reduce the serum ALT level of model rats, Qinggan Fang and Huoxue Fang has no significant effect; Qinggan Fang, Huoxue Fang, Qinggan Huoxue Fang can significantly reduce the serum AST levels in ALD rats, There was no significant difference between the two. (3) Qinggan Huoxue Decoction and its decomposed prescriptions all reduced plasma endotoxin levels in model rats, and there was no significant difference between the two groups. (4) Qinggan Huoxue Decoction and its decomposed prescriptions can inhibit the activation of KC in model rats, Huoxue Decoction is better than Qinggan Huoxue Decoction, and Qinggan Huoxue Decoction is better than Qinggan Decoction. Conclusion: Qinggan Huoxue Decoction and its disassembled prescriptions play a preventive and therapeutic effect on ALD, and its mechanism of action may be related to the reduction of plasma endotoxin levels and inhibition of KC activation.
