目的对乳胶凝集实验未能确定血清群的脑膜炎奈瑟菌菌株,用PCR方法鉴定其血清群。方法收集菌种库中未能鉴定血清群的11株脑膜炎奈瑟菌,提取核酸后经多重PCR1和多重PCR2,如无血清群条带则扩增cnl-1相似型、cnl-3,根据扩增片段的不同长度确定相应的血清群。结果经PCR1反应11株菌株均扩增出250 bp大小的脑膜炎奈瑟菌(Nm)条带,其中A群2株,B群1株,C群2株,W135群2株。经PCR2反应未扩增出条带。余下4株为cnl-1类群。结论多重PCR方法可用于鉴定脑膜炎奈瑟菌血清群,尤其是乳胶凝集实验不能确定的菌株,对流行病学研究和实验室诊断具有重要意义。
Objective To identify serogroups of Neisseria meningitides serogroups by latex agglutination test. Methods 11 strains of Neisseria meningitidis were collected from the strain library. Multiplex PCR1 and multiplex PCR2 were performed after nucleic acids were extracted. If no serogroups were detected, cnl-1 and cnl-3 were amplified. The different lengths of the amplified fragments determine the corresponding serogroups. Results A total of 250 isolates of N. meningitidis were amplified from 11 isolates by PCR1 reaction, including 2 strains of A group, 1 strain of B group, 2 strains of C group and 2 strains of W135 group. The PCR2 reaction did not amplify the band. The remaining 4 strains were cnl-1 groups. Conclusion The multiplex PCR method can be used to identify the serogroups of Neisseria meningitides, especially those strains that can not be determined by latex agglutination test. It is of great significance for epidemiological studies and laboratory diagnosis.
