目的研究大胡蜂窝乙醇抽提物(WCE)对60%酸性乙醇诱导的大鼠胃溃疡的保护性及其作用机制。方法用WCE按照0、25、100和150mg/kg的剂量对Wistar大鼠连续灌胃8d,然后用60%的酸性乙醇诱导胃溃疡,比较不同处理组大鼠胃粘膜的病变程度、髓过氧物酶活性和总抗氧化能力,同时分析WCE体外抗二苯基三硝基肼自由基的能力。结果与0mg/kgWCE灌胃组相比,25、100和150mg/kgWCE灌胃组的胃溃疡程度下降了44.2%~87.1%;胃粘膜髓过氧物酶活性下降了16.4%~56.6%,胃粘膜的总抗氧化能力分别增加0.5、1.47和1.83倍。此外,WCE体外自由基清除能力显著高于二丁基羟基甲苯。结论通过降低胃粘膜自由基的破坏性和中性粒细胞的渗漏性,WCE能显著抑制60%酸性乙醇诱导的大鼠胃溃疡和溃疡形成过程中的氧化反应。
Objective To study the protective effect of ethanolic extract of Magnolia cristata (WCE) on gastric ulcer induced by 60% acidic ethanol and its mechanism. Methods Wistar rats were continuously gavaged with 0, 25, 100 and 150 mg / kg for 8 days with WCE and then gastric ulcer was induced by 60% acidic ethanol. The pathological changes of gastric mucosa in different treatment groups were compared, Enzyme activity and total antioxidant capacity, while WCE in vitro anti-diphenyl trinitrohydrazide free radical capacity. Results Compared with the 0 mg / kg WCE group, the gastric ulcer levels in the 25, 100 and 150 mg / kg WCE groups decreased by 44.2% -87.1%; the gastric mucosal myeloperoxidase activity decreased by 16.4% -56.6% Total mucosal antioxidant capacity increased by 0.5,1.47 and 1.83 times, respectively. In addition, WCE in vitro free radical scavenging capacity was significantly higher than that of dibutylhydroxytoluene. Conclusion WCE can significantly inhibit the oxidative reaction in 60% acidic ethanol-induced gastric ulcer and ulcer formation in rats by reducing the destruction of gastric mucosal free radicals and neutrophil leakage.
