目的观察DEN诱发大鼠肝癌过程中C-myc、胰岛素样生长因子Ⅱ(IGF-Ⅱ)基因与细胞周期蛋白cyclinD1的表达变化及其相互关系。方法用原位杂交技术及免疫组化方法检测了大鼠肝癌形成过程中C-myc、IGF-Ⅱ mRNA及cyclinD1的表达变化,计算机图象分析检测结果,并进行统计学相关性分析。结果 C-myc、IGF-Ⅱ基因及cyclinD1在诱癌早期即可检出较多阳性信号,并随着大鼠肝癌诱发的进程逐渐增加,三者表达变化相关性分析具有显著意义(c-myc与IGF-Ⅱ基因阳性信号呈正相关(r=0.8125,P<0.001)。两基因与cyclinD1表达变化呈正相关(r=0.6607,P<0.001、r=0.6071,P<0.001)。结论 C-myc、IGF-Ⅱ基因过表达是肝癌发生的早期事件,并对癌细胞转化表型的维持具有一定意义。两基因的激活可能导致cyclinD1的过表达,三者的协同作用对肝细胞过度增殖以至癌变具有重要意义。
Objective To observe the expression and relationship of C-myc, insulin-like growth factor-II (IGF-II) and cyclinD1 during DEN-induced hepatocarcinogenesis in rats. Methods The expressions of C-myc and IGF-II mRNA and cyclinD1 were detected by in situ hybridization and immunohistochemistry. The results were analyzed by computer image analysis and statistical analysis was performed. Results The positive signals of C-myc, IGF-II and cyclinD1 were detected in the early stage of carcinogenesis, and gradually increased with the process of rat liver cancer induction. The correlation analysis between the three expressions was significant (c-myc). Positive correlation was found with positive signals of IGF-II gene (r=0.8125,P<0.001).The two genes were positively correlated with cyclinD1 expression (r=0.6607, P<0.001, r=0.6071, P<0.001).Conclusion C-myc, The overexpression of IGF-II gene is an early event of liver cancer, and has certain significance for the maintenance of the transformed phenotype of cancer cells.The activation of the two genes may lead to the overexpression of cyclinD1, and the synergistic effect of the three genes on the proliferation of hepatocytes and canceration.
