目的 :为了开展禽类重组IFN α类生物治疗制剂的研究。方法 :根据原始红鸡IFN A类基因设计了引物对 ,采用PCR法克隆了我国地方标准品种丝羽乌骨鸡的IFN α基因 ,并进行了序列分析 ;构建了鸡IFN α基因系统进化树。结果 :丝羽乌骨鸡的IFN α基因 (TSIFNA5 )与红色原鸡类鸡的IFN α基因相比 ,ORF长度都为 5 82个核苷酸、编码 193个氨基酸 ,相互间同源性在 97.4%~ 99.0 %。结论 :基因进化系统树揭示了TSIFNA5是鸡干扰素的一分支
Objective: In order to carry out the study of recombinant avian IFN class biological preparation. Methods: Primer pairs were designed according to the original class A gene of red chickens. The IFNα gene of silk chickens in our country was cloned by PCR and sequenced. The phylogenetic tree of chicken IFNα gene was constructed. Results: Compared with the IFNα gene of red chickens, the IFNα gene of silk feather silky chicken (TSIFNA5) had 5 82 nucleotides in length and encoded 193 amino acids with a homology of 97.4 % ~ 99.0%.
