目的对一只航空器截获的鼠类进行鉴定,确定其可能的来源地。方法提取芬兰入境航班捕获鼠(20150904)肝脏DNA,用PCR方法扩增细胞色素氧化酶Ⅰ(cytochromecoxidaseⅠ,COⅠ)和细胞色素b(cytochrome b,Cytb)基因并进行测序。将测序结果与GenBank中的相似鼠种DNA序列进行同源性分析。结果成功扩增到入境鼠肝脏的COⅠ和Cytb基因,遗传进化分析显示该鼠为小家鼠MUS种群,与北欧(芬兰)和中国(北方)小家鼠属于同一种群,但不能确定该小家鼠的具体来源地。结论形态学和分子生物学鉴定方法相结合是对截获病媒生物种群鉴定的有效方法。
Objective To identify the rodents intercepted by an aircraft and determine their possible origin. Methods The liver DNA of rat (20150904) was captured from a Finnish inbound flight, and the cytochrome b oxidase Ⅰ (COⅠ) and cytochrome b (Cytb) genes were amplified by PCR and sequenced. Homology analysis of the sequencing results with similar murine DNA sequences in GenBank. Results COⅠ and Cytb genes were successfully amplified in the livers of imported rats. The results of genetic evolution showed that the mice were MUS population of Mus musculus, belonging to the same group with Mus musculus in Northern Europe (Finland) and China (North) The specific origin of the mouse. Conclusion The combination of morphological and molecular biological identification methods is an effective method for identification of intercepted vector organisms.
