使用抗CD3单抗诱导新鲜分离的人外周血单个核细胞(PBMC)凋亡,同时观察细胞因子对其影响。18h后电镜发现处理组发生典型凋亡改变,DNA电泳出现典型阶梯状条带。TUNEL法流式细胞仪检测发现处理组凋亡发生率39.6%,显著高于对照组(<0.01)。IFN-7/TNF-a可显著增强抗CD3单抗诱导的凋亡。IL-1~IL-3、TNF-=对凋亡无明显影响。CsA可抑制抗CD3单抗诱导的凋亡,但加入IFN-y则可完全逆转这种作用。结果表明抗CD3单抗可诱导PBMC发生凋亡,IFN-Y、TNF-与抗CD3有协同作用。
Anti-CD3 monoclonal antibody was used to induce apoptosis of freshly isolated human peripheral blood mononuclear cells (PBMCs), and the effects of cytokines on them were also observed. After 18h, the typical apoptotic changes were found in the treated group, and typical ladder-like bands appeared in the electrophoresis of DNA. TUNEL flow cytometry found that the incidence of apoptosis in the treatment group was 39.6%, significantly higher than the control group (<0.01). IFN-7 / TNF-a significantly enhanced anti-CD3 mAb-induced apoptosis. IL-1 ~ IL-3, TNF- = no significant effect on apoptosis. CsA inhibited anti-CD3 mAb-induced apoptosis, but adding IFN-γ completely reversed this effect. The results show that anti-CD3 monoclonal antibody can induce apoptosis in PBMC, IFN-γ, TNF-and synergistic effect of anti-CD3.
