目的:研究hsa-mi R-127-3p(mi R-127)在宫颈癌组织中的表达水平及其与临床病理特征之间的相关性,探讨mi R-127对宫颈癌细胞株Hela及Siha迁移和侵袭的影响。方法:Real-time PCR法检测50对宫颈癌及癌旁组织中mi R-127表达水平,分析mi R-127表达与临床病理特征的相关性。细胞转染mi R-127 mimics及mimics negative control,采用划痕愈合实验检测细胞迁移能力,Transwell法检测细胞侵袭能力。结果:mi R-127在宫颈癌组织中的表达明显低于癌旁组织(P<0.05)。mi R-127表达与淋巴脉管间隙浸润相关(P=0.048),而与年龄、FIGO分期、病理类型、分化程度、病灶大小、浸润深度、有无淋巴结转移无关(P>0.05)。过表达mi R-127可抑制Hela及Siha细胞的迁移和侵袭能力。结论:mi R127在宫颈癌组织中低表达,并且抑制宫颈癌细胞株Hela及Siha的迁移和侵袭,可能与宫颈癌的发生发展密切相关。
Objective: To study the correlation between the expression of hsa-mi R-127-3p (mi R-127) and clinicopathological features in cervical cancer and to investigate the effect of mi R-127 on cervical cancer cell lines Hela and Siha Impact of migration and invasion. Methods: Real-time PCR was used to detect the expression of mi R-127 in 50 pairs of cervical cancer tissues and adjacent normal tissues. The correlation between mi R-127 expression and clinicopathological features was analyzed. The cells were transfected with mi R-127 mimics and mimics negative control. The cell migration ability was detected by scratch healing assay. The cell invasion ability was detected by Transwell assay. Results: The expression of mi R-127 in cervical cancer tissues was significantly lower than that in paracancerous tissues (P <0.05). The expression of mi R-127 was correlated with lymphatic vascular invasion (P = 0.048), but not with age, FIGO stage, histological type, degree of differentiation, lesion size, depth of invasion, and lymph node metastasis (P> 0.05). Overexpression of mi R-127 inhibited the migration and invasion of Hela and Siha cells. Conclusion: The low expression of mi R127 in cervical cancer and the inhibition of the migration and invasion of cervical cancer cell lines Hela and Siha may be closely related to the occurrence and development of cervical cancer.
