目的:观察鹿瓜多肽注射液对去势模鼠骨矿密度的影响。方法:将4月龄清洁级雌性SD大鼠40只随机分为假手术组(SHAM组),去势组(OVX组),去势+低剂量鹿瓜多肽组(OVX+CCP1组),去势+高剂量鹿瓜多肽组(OVX+CCP2组),每组10只。按照分组造模成功后,第13周起开始给药,分别为SHAM组:以0.9%NaCl肌注;OVX组:以0.9%NaCl肌注;OVX+CCP1组:以2mg/mL鹿瓜多肽注射液(0.4mL/kg/d)肌注;OVX+CCP2组:以2mg/mL鹿瓜多肽注射液(0.8mL/kg/d)肌注。用药频率均为每日1次,连续用药12周后断头法处死所有大鼠,采集腰4椎体和左股骨,测量骨密度。结果:用药12周后,SHAM组,OVX+CCP1组,OVX+CCP2组腰椎及左股骨骨密度均明显高于OVX组,差异有统计学意义(P<0.01);OVX+CCP1组,OVX+CCP2组骨密度均低于SHAM组,但差异无统计学意义(P>0.05);OVX+CCP2组骨密度高于OVX+CCP1组,差异有统计学意义(P<0.05)。结论:鹿瓜多肽注射液能够显著改善去卵巢后大鼠的骨密度,因此推论对雌激素水平低下的骨密度具有维持和改善作用;鹿瓜多肽在高低两种不同剂量下对改善骨密度均表现出差异性,显示剂量依赖趋势,提示鹿瓜多肽注射液可能具有剂量依赖效应。
Objective: To observe the effect of Lugua polypeptide injection on bone mineral density in castrated mice. Methods: Forty clean female Sprague-Dawley (SD) rats aged 4 months were randomly divided into sham operation group (SHAM group), castration group (OVX group), castrated + low dose Lugua polypeptide group (OVX + CCP1 group) Potency + high dose Lugua polypeptide group (OVX + CCP2 group), 10 in each group. The BMD of OVX + CCP2 group was higher than that of OVX + CCP1 group (P <0.05). Conclusion: Lugua Polypeptide Injection can significantly improve the bone density of ovariectomized rats, so we conclude that it has the effect of maintaining and improving the bone density with low estrogen level. Lugua polypeptide can improve bone mineral density both in high and low dosage Showing differences, showing a dose-dependent trend, suggesting that Lugua polypeptide injection may have a dose-dependent effect.
