目的:探讨雷公藤多甙对炎症性肠病(IBD)的治疗作用。方法:(1)动物模型:参照Morris法用30mgTNBS/50%酒精第一次致敏大鼠,并于三周后用10mgTNBS第二次致 敏,诱导复发。(2)治疗:实验设对照组、模型组、治疗组,在Morris法制成的模型上,给予雷公藤多甙4.5mg/kg/d35d,观察、比较各组大鼠大体和光镜下病理变化。结果:(1)动物模型:一次致敏的模型与Morris报道的病变基本一致,持续时间近五周,较Morris法短。用TNBS二次致敏未出现明显炎症和溃疡变化,但淋巴组织增生明显;(2)治疗:前7d一般情况与模型组相同,14d后活泼喜食,粪便性状良好,情况好于模型Ⅰ组,模型Ⅱ组;于治疗第三、四周时,模型组与治疗组比较光镜下病理改变显著减轻(P<0.05),于治疗第四、五周时,治疗组与模型组比较肉眼观察肠粘连有减轻的趋势,但无统计学差异。结论:用TNBS二次致敏大鼠制作复发性炎症性肠病模型的方法需进一步探讨;雷公藤多甙治疗实验性大鼠炎症性肠病有较好的疗效。
Objective: To investigate the therapeutic effects of Tripterygium wilfordii on inflammatory bowel disease (IBD). METHODS: (1) Animal model: The rats were sensitized with Morris method for the first time with 30 mg TNBS/50% alcohol and after 3 weeks with 10 mg TNBS for a second sensitization to induce relapse. (2) Treatment: The experimental group was divided into control group, model group, and treatment group. On the Morris method, the rats were given Tripterygium wilfordii Hg. 4.5 mg/kg/d for 35 days. The gross and light pathological changes of the rats in each group were observed and compared. Results: (1) Animal model: The primary sensitization model was basically consistent with the Morris reported lesion. The duration was nearly five weeks and shorter than the Morris method. Secondary sensitization with TNBS did not show significant inflammation and ulceration, but lymphoid hyperplasia was evident; (2) Treatment: The first 7 days were generally the same as the model group, 14 days later lively eating, fecal traits were good, the situation was better than the model I group At the third and fourth weeks after treatment, the pathological changes under light microscope were significantly reduced in the model group and the treatment group (P<0.05). At the fourth and fifth week of treatment, the treatment group and the model group were compared with the naked eye. There was a trend of reduction in adhesions, but there was no statistical difference. Conclusion: The method for making recurrent inflammatory bowel disease model in rats sensitized with TNBS should be further studied. Tripterygium wilfordii can effectively treat inflammatory bowel disease in experimental rats.
