参附注射液抗心源性休克大鼠ED_(50)的测定

目的:探讨心源性休克大鼠静脉注射参附注射液的半数有效剂量(ED50)。方法:SD大鼠64只,随机分为空白对照组、模型组、六个剂量给药组(1.25、2.5、5、10、20、25ml/kg),除空白对照组外,其余各组均采用结扎左冠状动脉法复制大鼠心源性休克模型,造模成功后采用多导生理记录仪记录实时血压值(MAP)及心率(HR),实验结束后心尖取血,测定血清中内皮素-1(ET-1)和一氧化氮含量(NO)。用GraphPadPrism5.0软件选择Sigmoidal dose-response模型对剂量与各指标值进行回归得半数有效剂量ED50。结果:剂量对数与血压峰值回归得方程Y=63.45+23.77/(1+10(0.7579-X)×5.546)、Y=359.4+114.5/(1+10(1.28-X)×2.994),相关系数R2分别为0.9619、0.9984,HillSlope分别为5.546、2.994,ED50分别为5.727、19.07ml/kg,呈现良好的“S”型曲线关系,血压峰值、心率峰值随剂量的增大而增大;剂量对数分别与ET-1的浓度、NO的含量进行回归得方程Y=8.514+6.276/(1+10(X-1.343)×1.336)、Y=25.25+7.94/(1+10(X-0.4001)×1.357),相关系数R2分别为0.4040、0.5342,HillSlope分别为-1.336、13.57,ED50分别为22.04、2.513ml/kg。结论:由血压值求得的ED50更科学合理,血压对应的ED50=5.727ml/kg,ED5%=3.368ml/kg,ED95%=9.738ml/kg。 Objective: To investigate the half effective dose (ED50) of intravenous injection of Shenfu injection in cardiogenic shock rats. Methods: Sixty-four SD rats were randomly divided into blank control group, model group and six dose groups (1.25,2.5,5,10,20,25ml / kg). The curves of blood pressure and heart rate peaked with the increase of dose ; The logarithm of the dose was regressed with the concentrations of ET-1 and NO, respectively, and the equation was Y = 8.514 + 6.276 / (1 + 10 (X-1.343) × 1.336), Y = 25.25 + 7.94 / (1 + 10 -0.4001) × 1.357). The correlation coefficients R2 were 0.4040 and 0.5342, HillSlope was -1.336 and 13.57 respectively, ED50 was 22.04 and 2.513ml / kg respectively. Conclusion: The ED50 obtained from the blood pressure value is more scientific and reasonable. The blood pressure corresponds to ED50 = 5.727ml / kg, ED5% = 3.368ml / kg, ED95% = 9.738ml / kg.
