目的 探讨层粘连蛋白 (L N)和基质金属蛋白酶(MMP)— MMP- 2、MMP- 9与其组织抑制因子 (TIMP)TIMP- 1在原发性人脑胶质瘤侵袭行为中的作用。 方法 利用免疫组织化学技术 SP法检测 L N和 MMP- 2、MMP- 9以及 TIMP- 1在正常脑组织、高低度恶性胶质瘤和脑内转移瘤中的表达情况 ;通过电镜观察胶质瘤中血管基底膜变化特征 ,并与免疫组织化学表达结果作对照比较。 结果 (1)正常脑组织和胶质瘤血管基底膜及血管内皮细胞上均有 L N的表达 ,而瘤细胞未见染色。 L N的表达强度与胶质瘤恶性度呈正相关 (P<0 .0 5 ) ,在高度恶性胶质瘤侵袭边缘的细胞间可见 L N的弱阳性表达 ;而脑内转移瘤则表现散在的瘤细胞浆膜阳性表达 ,基底膜及内皮细胞未见表达。(2 )正常脑组织中 ,MMP- 2阳性表达在血管基底膜及血管内皮细胞上 ,其他部位未见表达 ,而 MMP- 9均无表达。在胶质瘤中 MMP- 2和 MMP- 9阳性染色不仅见于血管基膜及血管内皮细胞 ,而且也见于侵袭边缘的瘤细胞。 70例人脑胶质瘤标本中 ,MMP- 2、MMP- 9的阳性表达率分别为 40 % (2 8/ 70 ) ,6 7.1%(4 7/ 70 ) ,差别具有显著性 (P<0 .0 5 ) ;MMP- 2、MMP- 9在低度恶性胶质瘤中阳性率分别为 14.3% (4 / 2 8) ,32 .1%(9/ 2 8) ,差别无显著性 (P>0 .0 5 ) ;MMP- 2、MMP
Objective To investigate the role of laminin (LN) and matrix metalloproteinases (MMPs) - MMP-2, MMP-9 and their tissue inhibitor of metalloproteinase (TIMP-1) in invasiveness of primary human glioma. Methods Immunohistochemical SP method was used to detect the expression of LN, MMP-2, MMP-9 and TIMP-1 in normal brain tissue, high-low grade glioma and brain metastases; electron microscopy in gliomas The changes of vascular basement membrane were compared with the results of immunohistochemistry. Results (1) The normal brain tissue and glioma vascular basement membrane and vascular endothelial cells were L N expression, and no staining of tumor cells.
