用MTT比色、AgNORs银集结合图像分析以及抗Ⅲ型胶原免疫组化技术观察并证实了γ-IFN和地塞米松(Dexa)对成纤维细胞生长的抑制作用。结果如下:(1)培养第3d,250×103U/L的γ-IFN和100mg/L的Dexa对成纤维细胞生长的抑制率分别为43%和33%;(2)照后3、5d两种抑制剂对正常、受照射以及5-HT刺激的受照成纤维细胞增殖(OD值示之)均显示出明显的抑制作用;(3)250×103U/L浓度的γ-IFN和150mg/L的Dexa对成纤维细胞AgNORs合成显示最强的抑制作用;其AgNORs颗粒数在5-HT组分别为对照值的58%和56%,而AgNORs3个定量参数的均值分别为对照值的51%和44%2(4)上述浓度的两种抑制剂对5-HT刺激的成纤维细胞Ⅲ型胶原合成也显示明显的抑制作用,在培养第3dⅢ型胶原阳性细胞数分别为对照值的62%和43%。
