本文在体外对新生大鼠(0~1d)大脑皮层神经细胞进行原代培养的基础上,建立谷氨酸(Glu)对培养的皮层神经细胞兴奋毒损伤模型。通过检测乳酸脱氢酶释出率和形态学观察,证实了Glu对神经细胞存在兴奋毒作用。通过测定不同条件下Glu兴奋毒对培养的神经细胞造成损伤时细胞膜脂质过氧化物(LPO)浓度,发现膜LPO浓度与Glu剂量及作用时间紧密相关,表明Glu对神经细胞兴奋毒作用有自由基产生并介导了神经细胞的损伤。
Based on the primary culture of neurons in the cerebral cortex of newborn rats (0 ~ 1d) in vitro, a model of excitotoxicity of glutamate (Glu) to cultured cortical neurons was established.
