一、引言为了鉴定种子园各无性系号及杂交后代优、劣,确定杂种优势,人工诱变多倍体等,除了进行表型分析以外,还要进行细胞学观察,主要是染色体的研究。一般都以发芽种子的根尖分生组织制成片子,进行树木染色体的研究。而需要研究的树木,很多还处于幼龄,无种子或种子很少,不够用。因此,取用生长旺盛的根尖或新针叶基部的细胞,制成片子进行染色体观察较方便。但大树挖根尖很困难,因此针叶芽染色体制片就更好些。关于针叶芽染色体制片技术,国外有过报道(1957年FRAN-COIS MERGEN,HELEN M. NOVOTNY曾研究过“湿地松针叶与根尖染色体研究的压挤
