通过观察外源性层粘素(LN)对一株新近建立的胆管癌细胞系QBC939分泌Ⅳ型胶原酶及体外浸润能力的影响,旨在探讨胆管癌细胞浸润过程中的生物学机理,特别是LN及Ⅳ型胶原酶在浸润中的作用和意义。结果表明,不同剂量的LN均明显刺激QBC939细胞分泌Ⅳ型胶原酶,且5μg/ml剂量的LN即能显著增强QBC939细胞的体外浸润能力。与LN同时加入LN抗体,显示不同剂量的LN抗体能不同程度地抑制LN对QBC939细胞Ⅳ型胶原酶分泌的刺激效应,并抑制LN增加QBC939细胞体外浸润能力的作用。研究结果显示,LN通过刺激癌细胞分泌Ⅳ型胶原酶,增强癌细胞浸润能力,在介导肿瘤细胞的浸润过程中发挥重要作用。Ⅳ型胶原酶在胆管癌浸润过程中起重要作用,其活性高低与癌细胞浸润能力相关,是分解细胞外基质的主要蛋白水解酶之一。研究结果不仅有助于癌浸润和转移机制的研究,同时也提示在未来抗肿瘤治疗中,干扰LN介导过程或抑制Ⅳ型胶原酶活性,有可能获得显著效果。
By observing the effect of exogenous laminin (LN) on the secretion of type IV collagenase and in vitro infiltration ability of a newly established cholangiocarcinoma cell line QBC939, the aim was to investigate the biological mechanism during the infiltration of cholangiocarcinoma cells, especially The role and significance of LN and type IV collagenase in infiltration. The results showed that different doses of LN significantly stimulated the secretion of type IV collagenase from QBC939 cells, and LN at a dose of 5 μg/ml could significantly enhance the in vitro infiltration ability of QBC939 cells. The simultaneous addition of LN antibody to LN showed that different doses of LN antibody could inhibit the stimulatory effect of LN on type IV collagenase secretion in QBC939 cells to varying degrees, and inhibit LN from increasing the in vitro infiltration ability of QBC939 cells. The results showed that LN plays an important role in mediating the infiltration of tumor cells by stimulating the secretion of type IV collagenase from cancer cells and enhancing the infiltration ability of cancer cells. Type IV collagenase plays an important role in the invasion of cholangiocarcinoma, and its activity is related to the infiltration ability of cancer cells. It is one of the major proteolytic enzymes that breaks down the extracellular matrix. The results of the study not only contribute to the study of the mechanisms of invasion and metastasis of cancer, but also suggest that in future anti-tumor therapy, interference with LN-mediated processes or inhibition of type IV collagenase activity may have significant effects.
