目的 观察精索静脉曲张模型大鼠睾丸组织凋亡相关基因Bcl-2、Fas与FasL的表达情况,以及补肾活血方对这些基因表达的影响。方法 将50只3月龄雄性SD大鼠先随机抽出10只为假手术组,其余40只大鼠采用肾静脉缩窄法建立精索静脉曲张病理模型,造模完成后再随机均分为模型组、补肾活血方低剂量组、中剂量组、高剂量组。造模完成后48d开始给药,连续给药60d,用免疫组织化学法检测各组大鼠睾丸组织凋亡相关基因Bcl-2、Fas、FasL的表达。结果 模型组大鼠睾丸组织Bcl-2蛋白表达下降,Fas、FasL蛋白表达上升;补肾活血方低、中、高剂量组睾丸组织Bcl-2蛋白表达显著上升,Fas、FasL蛋白表达显著下降。结论 补肾活血方可上调抑凋亡基因Bcl-2的表达,下调促凋亡基因Fas、FasL的表达,这可能是补肾活血方抑制生精细胞凋亡,改善精索静脉曲张性不育患者生精功能的机制。
Objective To observe the expression of apoptosis-related genes Bcl-2, Fas and FasL in the testicular tissue of varicocele rats and the effect of Bushen Huoxue Fang on the expression of these genes. Methods 50 male Sprague Dawley rats aged 3 months were randomly selected from 10 sham operation groups. The remaining 40 rats were established with a renal vein stenosis method to establish a varicocele pathological model. Group, Bushen Huoxue Fang low dose group, middle dose group, high dose group. The administration was started 48 d after the completion of the model, continuous administration for 60 days, and the expression of apoptosis-related genes Bcl-2, Fas, and FasL in the testis of each group was detected by immunohistochemistry. Results The expression of Bcl-2 protein in the testis of the model group decreased, and the expression of Fas and FasL protein increased. The expression of Bcl-2 protein in the testis of the Bushen Huoxuefang group increased significantly, and the expression of Fas and FasL protein decreased significantly. Conclusion Bushen Huoxue Decoction can up-regulate the expression of anti-apoptotic gene Bcl-2 and down-regulate the expression of pro-apoptosis gene Fas and FasL. This may be due to Bushen Huoxue Recipe inhibiting apoptosis of spermatogenic cells and improving varicocele infertility patients. The mechanism of fine function.
