  Zhikong scallop (Chlamys farreri) is one of the most important maricultural shellfish in the north of China.Although many genetic studies have been conducted for this species, little is yet known about its genome architecture.BAC libraries provide an excellent platform for gene isolation, creation of physical maps and complete genome sequencing.We constructed two bacterial artificial chromosome (BAC) libraries for Zhikong Scallop.The BAC libraries were generated based on two restriction enzymes, BamHI and HindⅢ, respectively.The BamHI library consists of 34,560 clones, with an average size of 98kb, covering 2.7 x haploid genome equivalents, while the HindⅢ library consists of 132,480 clones, with an average insert size of 106 kb, thus providing a genome coverage of 11.7 x.The combined libraries collectively contain a total of 167,040 BAC clones representing14.4 x haploid genome equivalents and having a probability of greater than 99% of discovering at least one positive clone with a single-copy sequence.BAC stability assays indicate that Zhikong scallop DNA was stable during propagation in the BAC system.The BAC libraries will serve as an invaluable resource for future genetic studies on Zhikong scallop such as positional cloning,physical mapping and even the large scale sequencing of Zhikong scallop genome.
