The t(8 ;21) (q22 ;q22) , representing the most common Chromosome translocation in AML, generates the fu sion oncoprotein AML1-ETO that is involved in leukemogenesis.Nevertheless, knock-in and transgenic mice stud ies were not demonstrated to develop any malignancies or leukemia. The study showed that "gain-of-function" mu tation and over-expression of C-KIT, encoding a receptor tyrosine Kinase, represents an important genetic event on the basis of AML-ETO, and cooperates with the AML1-ETO oncoprotein. These findings have not only enriched current knowledge of the pathogenesis of leukemia, but also provided important drug targets.
