The degeneration of dopamine neurons is a primary characteristic of Parkinsons disease.Although glial cell line-derived neurotrophic factor (GDNF) can greatly protect against dopamine neuron degeneration in the substantia nigra,it was not feasible for us to use this factor clinically because it cannot cross the blood brain barrier. However,it is important to explore the protective mechanism of GDNF on degenerative dopamine neurons. In this study,Sprague-Dawiey rats were divided into three groups: the control group, the 6-hydroxydopamine (6-OHDA) Parkinsons disease model group, and the 6-OHDA + GDNF group.Quantitative real-time polymerase chain reaction and western blotting were used to detect whether the transcription factor SIX2 participated in GDNF signaling in the substantia nigra. Results showed that mRNA expression of the SIX2 gene was decreased in the 6-OHDA group compared with the control group, and expression that in the 6-OHDA + GDNF group, the mRNA level increased compared with the 6-OHDA group. The changes in protein levels were consistent with the mRNA expression levels. These results suggest that the transcription factor SIX2 might be associated with GDNF signaling.
