大量的研究表明组蛋白乙酰化修饰在转录、DNA损伤修复和DNA复制等过程中起着重要作用.近年来研究表明除了乙酰化之外,组蛋白还能发生一系列类乙酰化修饰如丙酰、丁酰、巴豆酰等修饰.重要的是,一方面目前对催化这些修饰的酶体系了解的还较少,另一方面有证据表明CBP/p300不仅能够催化乙酰化,也能催化丙酰、丁酰和巴豆酰修饰.由于同一个酶可以催化不同的修饰,这一方面提出了之前归结于乙酰化修饰的功能是否可能是由于其它修饰所致,另一方面也为研究这些新的修饰的生物学功能设置了障碍.我们较系统的研究鉴定了哺乳动物细胞催化巴豆酰化及去除巴豆酰化修饰的酶体系,并构建了能够催化巴豆酰化但不能催化乙酰化的CBP/p300突变体,发现该突变体仍然具有促进转录的活性,表明巴豆酰化在转录调控过程中可能具有重要作用.
