  1.Background Melanin-concentrating hormone (MCH) is a cyclic 19-amino acid neuropeptide that is predominantly expressed in the central nervous system (CNS).It has been demonstrated that MCH plays an important role in the regulation of food intake and energy homeostasis.Two distinct sub-types of G-protein coupled receptors (GPCRs),MCH-1R and MCH-2R,are identified in mammals as specific receptors for MCH.The MCH-1R is widely expressed across many species while the MCH-2R was found in ferret,dog,monkey and human but not in rodents (e.g.,rat and mouse).Both rodent genetic and pharmacological evidences support that MCH-1R is a potential target for the treatment of obesity.Currently,GW856464,AMG-076 and NGD-4715 have entered clinical trials (Phase Ⅰ) for the treatment of obesity.
