Cyclic ADP-ribose(cADPR) is a second messenger molecule for mobilizing Ca2+in the endoplasmic stores via the ryanodine receptor in a wide range of cell systems spanning three biological kingdoms.It was discovered and its novel cyclic structure solved by us more than two decades ago.We went on to identify CD38,an ubiquitous protein with hitherto unknown function,is in fact the dominant enzyme in mammalian systems responsible for cyclizing the linear substrate NAD to produce cADPR,in a head-to-tail fashion with the release of the terminal nicotinamide group.We have delineated this novel cyclization reaction using crystallography and site-directed mutagenesis.Despite its importance,how this novel signaling enzyme is regulated remains largely unknown.In this presentation I will focus on the recent progress in elucidating the membrane topology of CD38 and the implications in understanding its mechanism of regulation.
