Translational research to move gene expression profiling from the research laboratory into the clinical arena is urgent in order to support clinical trials and improve patient care.Our CLIA-certified lab aims to analytically validate gene expression profiling for intrinsic subtype classification of breast cancer and apply it in clinical practice.Subtype-tailored treatments for Basal-like, HER2+, Luminal A & B are being developed, hence intrinsic subtype classification becomes important in clinical practice.We evaluated frozen breast cancer tissues by first using frozen section H+E to estimate the percentages of tumor cells, stroma/fat/blood, and necrosis.RNA was extracted from homogenized tissue by column purification and was evaluated for RNA integrity number (RIN) using an Agilent Bioanalyzer 2100.An Agilent Quick-Amp Labeling kit was used to generate tumor and reference cRNA labeled with Cy3 and Cy5, respectively.The labeled cRNAs was hybridized to an Agilent lx244K array overnight and then signals detected using an Agilent scanner.
