鞘脂信号通路与花粉管生长

  被子植物的有性生殖过程是植物生活的重要环节,与农作物产量形成密切相关.双受精过程的完成需要借助花粉管将精细胞运送到由珠被所包被的胚囊里面去,因此花粉管在体内的生长状况决定了最终的受精效率.花粉管在体内生长时,需要穿越雌蕊传导组织的细胞,同时也要感知并应答雌蕊及胚珠分泌的信号.我们通过正向遗传学的方法筛选得到影响雄配子体功能的突变体tod1((t)urg(o)r regulation (d)efect1),其花粉管内钙离子浓度下降,在花柱内生长缓慢,不能及时有效地从花柱道中穿出,导致部分雄性不育.膨压测定结果显示突变花粉管膨压增高,说明TOD1基因参与花粉管的膨压调控.TOD1基因编码一个定位在高尔基体的蛋白,它在花粉管和角果表皮的保卫细胞中特异性表达;生物化学实验证明TOD1具有碱性神经酰胺酶活性,它通过调节细胞内鞘氨醇及其下游活性分子——磷酸鞘氨醇的量和钙信号来调控细胞的膨压.上述研究表明,鞘脂信号通路在花粉管生长调控中具有重要的作用.
