Fluorine-19 nuclear magnetic resonance (19F NMR) spectroscopy is a powerful method in studying the conformation, topology and interactions of biological molecules.Here, we applied a 19F-containing amino acid (4-fluoro-L-phenylalanine, 4F-Phe) to label glucagon.With a standard curve for 4F-Phe chemical shifts for varying D20 concentrations, site-specific 19F solvent isotope shifts were analyzed for glucagon to compare solvent exposure of this peptide.
