Objective: To investigate the cytotoxic effect of the crude extract of S.barbata (CE-SB) on human hepatocarcinoma cells and further elucidate its effect on molecular mechanisms in MHCC97-H cells.Methods: 3-(4,5-dim ethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was used to examine the anti-proliferative effect of CE-SB.Apoptosis of MHCC97-H cells was observed under a fluorescence microscope with AO/EB staining and DNA ladder analysis.Mitochondrial transmembrane potential was determined using FCM with JC-1 staining.Protein level of Smac, Apaf-1, Caspase-9, and Survivin was measured by semiquantitive RT-PCR and Western blot analysis.The expression of the mitochondria-related proteins, Smac and Apaf-1, were also examined using immunofluorescence analysis.
