Objective: Glypican-3 (GPC-3), a membrane-associated heparan sulfate proteoglycan, is frequently up-regulated in hepatocellular carcinoma (HCC) and has proved that it is a valuable marker for HCC diagnosis and prognosis.Glypicans play a crucial role in the regulation of cell proliferation and survival, particularly during development and malignant transformation.Recent studies have revealed that GPC-3might regulate Wnts, Hedgehogs (Hhs) signaling pathway and fibroblast growth factors, bone morphogenetic proteins,involving in the development and invasion of HCC.In the case of Wnts, which are known to bind to heparan sulfate, overexpressed GPC3 promotes HCC growth by stimulating the canonical Wnt pathway.Wnt signaling pathway is highly conserved amonganimal species.Activation of the Wnt/β-catenin pathway controls a wide variety of processed in embryonic development and Mutations in adults promote cancer.Additionally, activation and accumulation of β-catenin is a critical event in Wnt/β-catenin signaling pathway regulation, the cell proliferation and survive.However, the mechanism of how the GPC-3 regulation the Wnt/β-catenin signaling in HCC cell is unclear.
