In our previous work,1 we synthesized PPET3-N2 and demonstrated its potential application in cell imaging.In the current work,the intracellular localization of PPET3-N2 was further studied.Cell imaging studies strongly displayed that PPET3-N2 can penetrate cell membrane and localize on lysosomes.Potential application of using the polymer as a lysosome stain was demonstrated to observe autophagy.Considering that terthiophene has high intersystem crossing and singlet oxygen sensitization efficiency,2-3 we also studied the ROS production ability of PPET3-N2.An interesting light-inducible translocation of PPET3-N2 in cells was observed.Upon light irradiation,lysosome was destructed due to the production of ROS.The polymer was then translocated to mitochondria.Furthermore,we also studied the phototoxicity of PPET3-N2 using MTT assay.The polymer displayed a prominent phototoxicity in HeLa cells with increasing light irradiation.
