Mutations in the Wnt/β-catenin signalling pathway, particularly truncations in the adenomatous polyposis coli (APC) gene, are the most common acquired genetic changes in sporadic colon cancer.Recent studies have also identified oncogenic mutations in B-Raf, K-Ras and PI-3-K (p 1 10) genes in colon cancer, and individual specimens often exhibit mutation in more than one gene.The relative contribution of these mutations to the tumor phenotype has not been investigated in detail; yet such knowledge would be invaluable in designing pathway-targeted therapeutic strategies.
