Hydroxyproline-rich glycoproteins (HRGPs) are reported to function in many cellular and physiological processes;however, their biological roles are not completely understood.In this study,we identified a novel Arabidopsis HRGP gene: NHRGP.Knocking out NHRGP did not result in obvious deleterious phenotypes, but transgenic plants overexpressing NHRGP (OE-N3 and OE-N6)exhibited several remarkable phenotypes in both vegetative and reproductive stages, including altered leaf and root morphology, inhibition of inflorescence elongation and thickening, defects in flower development and reduced seed production.Molecular characterization revealed that NHRGP was expressed differentially in organs and could be induced by various defense-related stimuli.Microarray and quantitative real-time PCR data showed that many defense-related genes as well as many cell wall modification genes were up-regulated in the OE-N6 compared with wild-type plants.Moreover, OE-N6 plants displayed enhanced resistance to Pseudomonas syringae L.pv tomato DC3000 in a pathogen infection assay.In sum, by means of phenotypic and bioinformatics analyses, we demonstrated that overexpression of NHRGP in Arabidopsis affects plant development and defense responses.
