Design: The secretome, a substance obtained from normoxia preconditioned human mesenchymal stem cells, can cause attenuation of traumatic brain injury (TBI) induced by fluid percussion injury (FPI) in animal model.A anaesthetised male Sprague-Dawley rats, immediately after the onset of fluid percussion TBI, were divided into two groups and given the vehicle solution (1 mL) or secretome (500μg) intravenously with sham group for control.Functional outcomes as motor (maximum grasp angle) by incline plane, cellular infarction volume by triphenyltetrazolium chloride (TCC) staining, and neuronal loss and apoptosis, neurotrophic factor as vascular epithelial growth factor (VEGF) expression in cortex by immuoflurocene staining were measured.All these parameters assessed on day 3 after traumatic injury.Results: Compared to those of sham-operated controls, the motor deficits and cerebral infarction of rats after TBI were significantly attenuated by secretome therapy.The TBI-induced neuronal loss and apoptosis are also significantly reduced by secretome therapy.Furthermore, VEGF-positive cells in the ischemic cortex after TBI were further significantly increased by secretome therapy.The results highlighted that intravenous injection of secretome from normoxia preconditioned human hMSC may ameliorate TBI-induced rats.We speculated secretome from normoxia preconditioned human MSC might be a promising therapy for traumatic brain injury in the future.
