A high-performance liquid chromatography coupled with electrospray ionization mass spectrometry method was presented for the simultaneous determination of five synthetic nonsteroidal anti-inflammatory drugs as well as two glucocorticoids,i.e.,piroxican,prednisone acetate,dexamethasone acetate,phenylbutazone,diclofenac,indometacin,and ibuprofen in twelve Chinese proprietary medicines for rheumatism.Glipizide was used as the internal standard.The separation was completed on a C18 column with the mobile phase consisted of methanol and ammonium acetate buffer (20 mM ammonium acetate and 0.1% formic acid) with gradient elution.The eight analytes were separated on baseline for preliminary qualitative assay and quantitative analysis.Full validation of the developed quantitative method was provided (linearity,limit of detection,limit of quantification,precision and accuracy) by HPLC-UV.LC-ESI-MS method was applied to further identify the structures of the analytes.The proposed method was suitable for safety assurance and quality control of suspect Chinese proprietary medicine for rheumatism.
