Objective: The paper investigated how exhaustive exercise and contusion affected satellite cells and myostatin (MSTN) in skeletal muscle of rats.Methods: 24 wistar male rats (7 weeks old) were randomly divided into four groups (n =6): the control group (C), immediately after exhaustive exercise group(E0), 24 hours after exhaustive exercise group (E24), 48 hours after exhaustive exercise group (E48).The rats of all exercise groups were given disposable treadmill exhaustive exercise and killed immediately, and at 24, 48 hours after exhaustive exercise.Then the left gastrocnemius muscle were removed and split in two.One was used for the cell culture in vitro immediately, and the other one kept in storage to liquid nitrogen to be measured for MSTN by ELISA.At the same time, 4 wistar male rats (7 weeks old) were arranged into two contusion groups(n=2): immediately after contusion instantly(D0), and 24 hours after contusion(D24).The left gastrocnemius of the rats was removed and used to culture the satellite cell in vitro immediately.On the other hand, the gastrocnemius muscles from two newborn rats (n=2) were immediately used for cell culture, too.
