DNA replication is a fundamental process conserved across species. The "Replicon" hypothesis, originally proposed by Jacob and Brenner,presented a basic concept for regulation of DNA replication.Subsequent genetic and biochemical studies in Escherichia coli provided molecular basis for the model.Plasmids.Extra chromosomal elements in bacteria,played important roles in elucidation of mechanisms involved in initiation and its regulation of replicons.My studies in plasmid Rl provided a unique model for "once and only once replication" by its initiator RepA protein.DNA chain elongation is initiated by primer RNA synthesis.I have shown the presence of multiple modes of primer RNA synthesis in E.coli,some of which are hijacked by non-pathogenic phages that are hosted in E.coli.PriA,capable of assembling a protein complex competent for primer RNA synthesis on the lagging strands.Can bind to stalled fork structures by specifically recognizing the 3-end of the stalled nascent leading strand.Genetic and biochemical studies indicate that PriA plays crucial roles in detecting a stalled fork and reinitiation of DNA replication,a process essential for maintenance of genome integrity.
