花香是花卉的一个重要的观赏品质,它在吸引昆虫授粉和繁育等方面具有重要的作用,并且具有节律性释放特性,但是对于植物如何有效地控制花香代谢网络来高效地吸引传粉者还不清楚.百合是世界重要的观赏植物,目前生产上主栽的东方百合、OT系百合和麝香百合品种均具有节律性挥发香气的特征.本研究采用能节律性挥发强烈香味的西伯利亚百合花瓣为材料,进行了转录组测序和挥发量高时间点(白天16:00)和低时间点(晚上24:00)的表达谱分析,通过Trinity软件拼接共获得29,837条高质量的Unigenes.上调基因(白天16:00)主要集中在萜类骨架合成、苯基苯丙烷代谢、糖和淀粉代谢,而下调基因(夜晚24:00)主要富集在α-亚麻酸代谢、糖和淀粉代谢.通过BLASTX比如Unigenes得到了36条与花香、节律、糖代谢相关的调控基因.通过荧光定量PCR分析4个相关基因在60h内表达量的节律变化.结果显示在白天节律钟相关基因FKF1和GI是上调基因,但CK2β1、CK2β2是下调基因,并且糖和淀粉代谢以及α-亚麻酸代谢基因是下调基因,但是在夜晚结果相反.这些数据将会对于百合花香节律变化和昼夜节律研究提供帮助.
