Fine genetic and physical mapping of the CRb gene conferring resistance to clubroot disease in Brass

  The use of clubroot resistance(CR)genes is an effective and economical approach to control Plasmodiophora brassicae,the causal agent of clubroot disease in Brassica rapa and other Brassica crops.In the previous study,we identified and mapped the CRb locus on chromosome A03 of B.rapa in Chinese cabbage doubled haploid line CR Shinki.The CRb gene,a dominant gene resistance to pathotype 4 of P.brassicae,was finely mapped in combination of bulked segregant analysis(BSA)and bioinformatics analysis(BIA).Using 1,486 highly susceptible individuals and 2,896 individuals from two separate F2 populations of CR Shinki x 702-5(a Packchoi DH line),the CRb locus was narrowed down to a region of about 0.14 cM between two flanking markers TCR79 and TCR108.To construct the physical map of this locus,the sequences of 7 newly developed markers and other polymorphic markers linked to CRa and CRbKato were landed on chromosome A03 in silico by BIA.The CRb locus was defined to about 83.5 kb interval of A03 chromosome on B.rapa reference genome,and this interval contains two Toll-interleukin-1 receptor/nucleotide-binding site/leucine-rich repeat(TIR-NBS-LRR)gene and several putative regulation genes in the B.rapa genome.Fine genetic and physical mapping indicated that CRb is a different CR gene compared with previous mapped CRa and CRbKato by other researches.The results obtained in here provide useful information for isolation of the CRb gene and tightly linked molecular markers for CR breeding in B.rapa.
