Objective:Via rat spinal cor injury model,design and screen the optimized RNA interfere molecular targeting the gene coding region of rat IL-6 receptor α subunit,and construct adenovirus expression vector.Methods:Three pairs of shRNA template which composed of 19 bp reverse repeated motif of IL-6 receptor α target sequence with 9 bp spacer were designed and synthesized,then the adenovirus recombinants with red fluorescence protein gene were constructed respectively.The acute spinal cord injury model was completed,then the adenovirus recombinants were regional injected post 12h after SCI; the injuryed regional expression level of IL-6R were detected via real time fluorescence quantitative RT-PCR and Western blot; the optimal shRNA adenovirus recombinants of expression suppressive of IL-6 receptor α was screened.
